Waiguru attended Nairobi River Primary school and later she joined Precious Blood Girls’ Secondary school Riruta and sat for her O-levels in 1987. She proceeded to Moi Forces Academy and did mathematics, physics and chemistry. She did her A-levels in 1989.
Waiguru later joined Egerton University where she graduated with a bachelor’s degree in agriculture and home economics. She is also a holder of master’s degree in economics from the university of Nairobi with specialization in public finance, financial management systems, public service reforms, capacity building and governance. She also studied at Kenya Leadership Institute.

Ann Waiguru worked briefly as an intern at the Transparency International after finishing her degree education at Egerton university.
Her career in public service began when she worked at the Public Service Reform Secretariat where she provided technical service. She the served as the head of governance and the economic stimulus program at National Treasury.
Waiguru was also the director of Integrated Financial Management and Information System and head of governance at national treasury. Uhuru Kenyatta appointed her as Cabinet secretary in the ministry of Devolution where she was later entangled in the NYS scandal that saw billions lost. This tainted her image.
In 2016, she expressed her interests in politics and she joined the TNA party in readiness to vie as governor for Kirinyaga in the 2017 elections. She was successfully elected as the second governor of Kirinyaga and she defended the seat in 2022 election through UDA party.

She remained single for a long time until her traditional wedding at Kamagumo primary school in 2019 to lawyer Kamotho Waiganjo whom they had been friends with for over ten years.
She has three sons named Don, Ian and Wabu. It is not clear whether the children she got with her first husband Tony Waiguru Njuguna whom they broke up over infidelity allegations.
